ソフトウェアThorImageLSは、当社の顕微鏡およびその他の補助機器の動作を制御して(詳細は「仕様」タブ参照)データセットの取得および解析を行います。当社で開発されたバージョン4.3には、ホログラフィックオプトジェネティクス、生体深部スキャン、広域タイリング、高速イメージング、マルチモ-ダル画像の取得といった様々なイメージング手法を実行するための機能がございます。ThorImageLSの機能の詳細については、下の画像または上のタブをクリックしてご覧ください。

ThorImage®LSソフトウェアのワークスペース

ThorImageLSを用いると、その直観的で多機能なワークフローにより、画像の取得および解析プロセスを効率化することができます。コントロールパネルは重要な機能にすぐにアクセスできるように設計されており、実験のセットアップを素早く行うことができます。パネルのレイアウトは全てカスタマイズ可能で、様々なユーザやイメージング手法に対応することができます。全てのソフトウェア機能へのアクセスが容易な、操作性に優れた自己完結型のパッケージ型ソフトウェアです。

5D可視化

ThorImageLSは完備されたソフトウェアパッケージとしてご提供しており、すべてのXYZCTデータセットを取得できます。X、Y、Z方向の3Dボリューム画像を取得し、それを経時的に追跡して動的な事象を調べることができます。同じソフトウェアの中で、複数の蛍光色素画像や複数のイメージング手法による画像を重ねることで、5D情報をそれぞれ独立に追加することができます。セットアップおよび画像取得は、データ量が多い場合でもシンプルかつ直観的に行えるように設計されています。

顕微鏡での複数のイメージング手法を利用した画像取得を簡素化するために、ThorImageLSは以下のイメージング手法に対応しています。

1つのソフトウェアパッケージでマルチモーダル画像を取得でき、複数のソフトウェアアプリケーションを切り換える必要がありません。レーザ走査イメージとワイドフィールドイメージを同時に取得可能です。それに加えてThorSync™を使用すれば、外部カメラのデータと顕微鏡の画像データを同期させて、画像の特徴と試料の動きとの相関を見ることもできます。

連続画像取得

ThorImageLSは、顕微鏡とレーザ光源の動作のタイミングを正確に調整して、標準モードまたは高速モードで連続して画像取得ができます。当社のマルチチャンネル共焦点システムを用いると、登録された画像や画像スタックを取得するために波長を簡単に切り替えることができます。また、当社のTIBERIUS®フェムト秒Ti:サファイアレーザを用いるとマルチチャンネルによる高速な多光子イメージングが可能になり、信号対雑音比(SNR)が高くチャンネル間クロストークの小さい画像が得られます(右図がその一例です)。

光活性化
オプトジェネティクスのための光活性化やプロテインのアンケージングは、ThorImageLSを用いると使用機器に合わせて様々な方法で行うことができます。

Bergamo®IIシステムに対応する当社の空間変調モジュールはThorImageLSで直接制御でき、数百の関心領域(ROI)を同時に、個別に、またはグループごとに活性化することができます。右の画像は当社のソフトウェアを用いて顕微鏡の視野内にある100個のニューロンを特定し、それらに同時刺激を与えた様子です。

ガルバノ-ガルバノスキャナを使用して、何千もの関心領域を順次連続的に刺激することもできます。細胞全体を確実に刺激できるように、ガルバノ走査パターンと関心領域のタイプを指定できます。この活性化は画像取得と正確に同期しており、高感度なPMTの寿命を延ばすことができます。

全視野刺激を行う場合は、LEDまたはレーザ光源のどちらかを使用します。刺激は画像取得の前または後、あるいは各フレーム間でも行うことができます。

マスキング
ガルバノ-レゾナントスキャナ方式のレーザ走査顕微鏡の場合には、位置ごとのレーザ光の滞在時間が一様でなくなり、特に走査領域の末端においてそれが顕著に現れます。ThorImageLSはポッケルスセルを制御して(Bergamo IIシステムで使用可能)エッジの消去および関心領域の高速マスキングを行うことで、領域内のレーザ励起の均一性を向上させるとともに、領域外の不要な露光を防止します。関心領域はライブ画像内を単にドラッグ&ドロップするだけで指定できます。

高速ビデオ

画像セットは最大400 fpsで取得できますが、保存可能な容量は対応するハードドライブのサイズよって制限されます。ボリューム画像取得時は、顕微鏡のピエゾによる高速なZ方向移動量とデータ取得を同期させ、ユーザ定義のステップサイズで正確に走査することができます。データの取得途中でも、そのビデオフレームレートでそれまでに取得された画像セットあるいはストリーミングの画像セットに対して、関心領域のリアルタイム解析を行うことができます。そのため高感度なin vivoの実験における停止時間を短縮できます。

生体深部スキャン

ThorImageLSは補助的なハードウェアの動作を調整して、深部の高速3D画像スタックをより効率的に取得できます。移動距離の長いステッピングモータや高精度のピエゾモータを用いて、ユーザが定めたステップサイズでZ軸に沿って広範囲に移動させることができます。組織の深さに依存する光散乱の影響は、深さに応じてレーザ光のパワーを変化させて補正することができ、走査中の信号応答を均一化できます。

関心領域のリアルタイムでの解析および画像修正

画像取得と同時にリアルタイムで演算処理ができるように、画像の取得前または取得中でも関心領域の選択ができます。関心領域は様々な形状で選択でき、光活性化においては離れた集団をグループ化することもできます。関心領域について以下の計算が可能です:

  • 強度の平均値、最小値および最大値
  • 標準偏差
  • 数値演算
    • dF/F
    • レシオメトリックイメージング

広域のモザイクタイリング

関心領域を観察するのに1回の画像走査では十分な分解能が得られない場合は、試料面を連続したタイルのように画像化していくことができます。それらをソフトウェアでスティッチング(stitch、縫合)し、1枚の高分解能な合成画像にします。タイルの位置、サイズ、およびタイルのオーバーラップの設定、さらに試料と対物レンズとの非直交を修正するためのチルト調整が可能です。この機能は、時系列の画像セットと3D画像スタックの両方にも対応しています。

ThorSync™

ThorSyncは、デジタル信号とアナログ信号を最大2 MHzの速度でモニタし保存します。このソフトウェアアドオンはThorImage®LSと連携して動作するように設計されており、顕微鏡のデータ取得と補助的なハードウェアを連携させその処理プロセスを同期させます。ThorImageLS同様に、ThorSyncのアプリケーションコードもご提供しているので、ソフトウェアのフルカスタマイズが可能です。

このソフトウェアパッケージは、光励起/光刺激などの実験を開始したり調整したりするために、任意のデジタル波形を出力します。デジタル信号やアナログ信号のサンプリングには同じベースクロックが使われており、得られたデータはPCのハードドライブに保存されます。データサンプリング用に、アナログ入力チャンネル(AI)が16、デジタルTTL入力チャンネル(DI)が32あります。アナログ信号は+/-10Vに対して16 bitの分解能でサンプリングできます。光とシャッタの制御用デジタル出力信号(DO)のリフレッシュレートは最大2 MHzです。このソフトウェアは、顕微鏡のメインPCに組み込まれたPCIeベースのDAQを利用します。

ThorSyncは、データ取得中および取得後に、可視化された詳細な波形情報を保存します。ロールオーバ統計によりすべての信号のリアルタイム情報が提供され、ユーザが各波形内の位置を指定することで時間とY軸の値を表示できます。ThorImageLSと同様に、取得できるデータセットの大きさは、保存先として選択されたハードドライブのサイズにのみ制限されます。ThorSyncのデータは、ほとんどのデータ解析パッケージで容易にインポートできる一般的なHDF5形式で保存されます。

サードパーティソフトウェアとの統合による自動取得および解析

ThorImageLSでデータ取得から解析までを自動化するには様々な方法があり、ThorImageLSを単独で使用することもサードパーティのソフトウェアやコードと連携させることもできます。

ThorImageLSのスクリプト
画像取得の設定とコマンドのシーケンスは、スクリプト作成エリアに直接ドラッグ&ドロップすることで作成できます。このインターフェイスによりプログラミングの深い知識は必要ありません。また、ImageJのマクロおよびMATLABのスクリプトは、スクリプトエリアからスクリプトコマンドで直接実行できます。スクリプトはXMLファイルとしてエクスポートでき、別のユーザやワークステーションとシェアできます。

サードパーティとの統合
当社のソフトウェアは、MATLABおよびFiji/ImageJを用いて画像取得と解析の自動化を容易に行うことができます。取得したデータセットは、ThorImageLSソフトウェア内のquick connectボタンを使用して、MATLABまたはFiji/ImageJで直接開くことができます。このソフトウェアでMATLABスクリプトとFiji/ImageJマクロを直接実行することもできます。

データエクスポートの特徴
画像データおよび補助データは一般的なファイル形式で保存されるため、他のソフトウェアとの統合化が容易です。画像はメタデータに保存された校正値と一緒に16ビットのTIFFまたはRAWファイルとして保存されます。補助機器および実験条件の設定内容はXML形式で保存されます。ThorSyncと関心領域の統計データはHDF5フォーマットで保存されます。

ソフトウェア開発キット(SDK)
ThorImageLSには、お手持ちのデバイス用プラグインのコードを作成するためのオープンソースSDKがございます。「オープンソース」タブ内のフォームで当社にご連絡いただければ、SDKやソフトウェアの全コードにアクセスすることができます。

ThorImage®LS用コードへのアクセス

研究コミュニティのニーズにお応えして、ThorImageLSおよびそのアドオンのThorSyncはオープンソースとし、対応する当社の顕微鏡システムに付属しています。プログラムはMicrosoft C#およびC++で書かれており、お客様のニーズに合わせてフルカスタマイズすることが可能です。当社のソフトウェアについては長期にわたるサポートをご提供しております。製品の詳細情報、あるいはGitLabにアクセスして最新のThorImageLS(バージョン4.3)やThorSyncソフトウェアのソースコードをダウンロードする方法については、以下の様式で当社までお問い合わせください。お客様からのご提案やバグに関するレポートもお待ちしております。ThorImageLSバージョン3.0およびそれ以降のバージョンは、バージョン2.xおよび 1.xに比べて大幅に新機能が追加されているため、バージョン3.0およびそれ以降のバージョンは当社の旧モデルの顕微鏡には対応しておりませんのでご注意ください。当社では旧モデルをお持ちのお客様のために旧バージョンのソフトウェアのサポートを継続しております。

Hi Bergamo team, we are running Thorimage on our Bergamo and the system was initially setup with a resonant galvo. We now want to switch to a galvo-galvo system. Is there an easy way to change this hardware change in the Thorimage software? Thanks!

您好,我想請問幾個問題。 ThorImage®LS這套軟體,是提供購買您們產品的客戶免費下載的嗎?還是需要另外再購買呢? 官網上介紹的工作區介面,是軟體自帶的嗎?還是用戶必須自己去寫呢? 我們主要是想通過「ThorImage®LS→DAQ→Galvo Scanning System」再去掃描sample,在ThorImage®LS上面顯示galvo scanning system掃描到的成像圖,不知道實際上能不能這樣操作....? 謝謝。

Thank you for contacting Thorlabs. At the moment our Thorimage software is only available when included with one of our imaging systems and we do not have a version of the Thorimage software to download separately for DIY projects. Access to the open source project is limited to existing customers looking to customize Thorimage to their needs with Thorlabs microscope. The scanning parts Thorimage is not open source and cannot be customized to function with other scanners and digitizing hardware.
